Diana Agrest führt seit 1980 zusammen mit Mario Gandelsonas das Büro Agrest and Gandelsonas Architects in New York, hat zudem ein eigenes Büro und lehrt an der Cooper Union. Von 1972 bis 1984 war sie Mitglied des Institute for Architecture and Urban Studies, und leitete dort den Advanced Workshop in Architecture and Urban Form. Sie ist Autorin diverser Monographien, u.a. von „Architecture from Without. Theoretical Framings for a Critical Practice“ (1991), „The Sex of Architecture“ (1996) und „On Practice. An Architecture of Criticism, Production of Place, the Mirror of Enigams“ (2000). Gegenwärtig arbeit sie an einer Dokumentation zum IAUS mit dem Titel „The Making of an Avant-Garde“.
